** AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** THIS PROJECTRESPONDS TO USDA-NIFA AFRI FOUNDATION PROGRAM FOR FOOD SAFETY, NUTRITION AND HEALTH IN THE PROGRAM AREA OF FOOD AND HUMAN HEALTH (A1343). WE WILL EXAMINE INTERACTIONS BETWEEN BRASSICA, GUT MICROBIOME (GM) AND GLP-1 SECRETION. BRASSICA VEGETABLES ARE A RICH SOURCE OF GLUCOSINOLATES (GSLS), A GROUP OF SULFUR COMPOUNDS THAT ARE UNIQUE TO THESE VEGETABLES (BROCCOLI, KALE, AND OTHERS). UPON HYDROLYSIS, GSLS ARE CONVERTED TO BIOACTIVE ISOTHIOCYANATES (ITCS). WHEN CONSUMED RAW, GSLS CAN BE CONVERTED TO ITCS BY THE PLANT ENZYME MYROSINASE. HEAT (COOKING) INACTIVATES MYROSINASE, THEN GUT MICROBIOTA (GM) BECOME RESPONSIBLE FOR ANY GSL CONVERSION TO ITCS. WE HYPOTHESIZE THAT NON-ITC PRODUCING PATHWAYS PRESENT IN THE GM ARE COMPETING FOR GSLS AND THUS EXPLAIN THE DRAMATIC INTER-INDIVIDUAL DIFFERENCES SEEN IN ITC PRODUCTION (AIM 1). ONCE THOUGHT TO BE ONLY PRESENT IN THE MOUTH, BITTER-TASTE RECEPTORS (T2RS) ARE NOW FOUND TO BE PRESENT IN MANY TISSUES INCLUDING THE GUT. SINCE GUT T2RS CAN TRIGGER GLP-1 SECRETION AND ITCS CAN SERVE AS T2R AGONISTS, WE HYPOTHESIZE THAT ITCS PRODUCED BY THE GM BIND INTESTINAL T2R(S) TO TRIGGER GLP-1-DEPENDENT CONTROL OF OBESITY AND GLUCOSE HOMEOSTASIS (AIM 2). OPTIMIZING ITC PRODUCTION IN THE GUT, BOTH AMOUNT AND LOCATION, HAS THE POTENTIAL TO REVERSE GLUCOSE INTOLERANCE AND OBESITY IN HUMANS. OUR FINDINGS WILL INFORM THE PUBLIC ABOUT THE HEALTH PROMOTING PROPERTIES OF BRASSICA, EXPLAIN THE VARIATION IN GSL BIOACTIVATION SEEN IN HUMAN STUDIES, WHERE NON-ITC METABOLISM OF GSLS BY GM HAVE BEEN LITTLE CONSIDERED, AND WILL FURTHER INCREASE BRASSICA INTAKE WORLDWIDE.

** AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** CURRENTLY, AMERICANS ARE CONSUMING 42% OF THEIR DAILY CALORIES FROM LOW-QUALITY CARBOHYDRATES, SUCH AS REFINED GRAINS AND SUGAR SWEETENED BEVERAGES. EXCESSIVE INTAKE OF REFINED CARBOHYDRATESIS A MAJOR DIETARY RISK FACTOR FOR THE DEVELOPMENT OF NONALCOHOLIC FATTY LIVER DISEASE (NAFLD) WHICH IS MORE PREVALENT AND SEVERE AMONGST AGED INDIVIDUALS. WHILE CRITICAL TO RECOMMEND DECREASED CONSUMPTION OF REFINED CARBOHYDRATES, IT IS ALSO IMPORTANT TO IDENTIFY FOOD-DERIVED, BIOLOGICALLY ACTIVE COMPOUNDS, SUCH AS LYCOPENE (A RED PIGMENT) AND PHYTOENE (A PRECAUSOR OF LYCOPENE AND OTHER MAJOR CAROTENOIDS)FROM TOMATOES AND TOMATO PRODUCTS, WHICH MAY DELAY THE ONSET OF NAFLD THROUGH TARGETING THE GUT-LIVER INTERACTION. IN THE PROPOSED STUDY, WE WILL EXPLORE WHETHER AGE (YOUNG MICE AT 8 MONTHS VS. OLDER MICE AT 24 MONTHS) AND CAROTENOID BREAKDOWN ENZYMES (NORMAL MICEVS. MICE LACKING CAROTENOID BREAKDOWN ENZYMES) MAY DIFFERENTIALLY CONTRIBUTE TO HIGH REFINED CARBOHYDRATE DIET (HRCD)-INDUCED NAFLD DEVELOPMENT; AND WHETHER TOMATO POWDER (WHOLE TOMATO) AND KEY TOMATO-BASED CAROTENOIDS (LYCOPENE AND PHYTOENE) ELICIT PROTECTION AGAINST HRCD-RELATED NAFLD THROUGH MODULATING THE KEY GENES (SIRTUIN 1 SIGNALING), GUT BACTERIA PROFILE (GUT MICROBIOME) AND METABOLIC PROFILE (METABOLOMICS)IN YOUNG AND OLDER ANIMALS. WE EXPECT THAT TOMATO POWDER, LYCOPENE, AND PHYTOENE INTERVENTIONS WILL DIMINISH HRCD-, AGE-RELATED ABERRANT GENE EXPRESSION, INFLAMMATION AND OXIDATIVE STRESS, CONSEQUENTLY MINIMIZINGGUT BACTERIA AND METABOLIC ALTERATIONS AND REDUCING NAFLD PROGRESSION. THESE DATA WILL PROVIDE NEW INSIGHT INTO THE PROTECTIVE EFFECTS OF TOMATO AND TOMATO PRODUCTS AND WILL FURTHER EMBOLDEN THE PREVENTIVE ROLE OF TOMATO AGAINST HRCD-ASSOCIATED NAFLD AND METABOLIC DISORDERS IN AGED POPULATION.

WITHIN THIS CONTEXT THE QUESTION HAS ARISEN AS TO WHETHER THE CURRENT GUIDANCE REGARDING DAIRY PRODUCTS FOR HEART DISEASE RISK REDUCTION SHOULD BE MODIFIED. TO CLARIFY THIS ISSUE A STUDY WAS DESIGNED TO COMPARE THE EFFECTS OF FULL-FAT AND FAT-FREE DAIRY, WITH AND WITHOUT FERMENTATION (YOGURT), ON HEART DISEASE RISK FACTORS AND DETERMINE IF THE EFFECTS OBSERVED ARE MEDIATED BY THE BACTERIA IN OUR INTESTINE (GUT MICROBIOME) AND THE COMPONENTS THEY PRODUCE. TO ACCOMPLISH THIS WE WILL CONDUCT A RANDOMIZED-CONTROLLED CROSS-OVER TRIAL COMPARING THE EFFECT OF CONSUMING TWO SERVINGS/DAY OF FULL-FAT MILK, FAT-FREE MILK, FULL-FAT YOGURT AND FAT-FREE YOGURT ON HEART DISEASE RISK FACTORS (E.G., BLOOD LIPID AND GLUCOSE CONCENTRATIONS) AND THE GUT MICROBIOME COMPOSITION AND ITS PRODUCTS. HEART DISEASE AND RELATED DISORDERS ARE THE LEADING CAUSE OF DEATH AND DISABILITY IN THE U.S. POOR DIET QUALITY IS A MAJOR CONTRIBUTOR. IT IS MORE CRITICAL THAT EVER TO HAVE THE EVIDENCE BASE ON WHICH TO FORMULATION THE BEST ADVISE.

THE DISEASE RISK INCREASES WITH AGE AND AFFECTS WOMEN MORE THAN MEN. NO OPTIONS YET EXIST TO REDUCE THE RISK OF EARLY AMD, BUT THE DIETARY CAROTENOIDS LUTEIN AND ZEAXANTHIN, WHICH ARE CONCENTRATED IN THE MACULA, SHOW PROMISE, SINCE THEY BLOCK DAMAGING BLUE LIGHT AND MODULATE OXIDATIVE STRESS. GOJI BERRIES CONTAIN THE HIGHEST AMOUNT OF ZEAXANTHIN AMONG ALL KNOWN DIETARY SOURCES, AND ALONG WITH THE LUTEIN AND A DISTINCTIVE POLYSACCHARIDE, MAY OFFER AN APPROACH TO REDUCE THE RISK OF AMD. WE HAVE RECENTLY SHOWN THAT GOJI BERRY INTAKE FOR 90 DAYS SIGNIFICANTLY INCREASED A BIOMARKER OF LUTEIN AND ZEAXANTHIN IN THE MACULA IN HEALTHY ADULTS. HERE WE PROPOSE TO ASSESS THE EFFECTS OF GOJI BERRY INTAKE FOR 180 DAYS AMONG PARTICIPANTS AT-RISK FOR AMD, AND COMPARE THEIR MULTIPLE OCULAR OUTCOMES TO A CONTROL GROUP CONSUMING CALORIE- AND FIBER-MATCHED GUMMIES OUR STUDY WILL ALSO PROFILE GUT MICROBIOME CHANGES USING SHALLOW SHOTGUN METAGENOMIC SEQUENCING, ASSESS DIFFERENCES IN ASSOCIATED PLASMA MICROBIAL METABOLITES, AND FOLLOW PLASMA LEVELS OF LUTEIN AND ZEAXANTHIN. THIS THREE-YEAR RANDOMIZED CLINICAL TRIAL ADDRESSES THE AFRI PROGRAM AREA PRIORITY FOOD AND HUMAN HEALTH USING A WHOLE FOODS APPROACH. IF SUCCESSFUL, THE RESULTS CAN GIVE CONSUMERS AND HEALTH CARE PROVIDERS NEW FOOD-BASED OPTIONS TO REDUCE THE RISK OF AMD AND PROMOTE BETTER EYE HEALTH. THE RESULTS MAY ALSO STIMULATE DEVELOPMENT OF GOJI BERRIES AS A VIABLE SPECIALTY CROP IN THE UNITED STATES.
